Comments
Topoisomerase II relieves DNA torsional stress in replication and transcription. Topoisomerase IIα and IIβ are the two isoforms in humans with 78% identity. Topo IIα and β are both targets for anticancer drugs. Mirski et al (BBRC, 2003) found that 1018-1028 of Topo IIα could target GST-GFP out of the nucleus. Mirski et al. 2007 further showed that recombinant Topoisomerase IIα bound recombinant CRM1 in GST-pulldown assay in the presence of RanGTP, but the protein does not shuttle between the nucleus and the cytoplasm in proliferating cells (heterokaryon shuttling experiments). Turner et al. found another NES (residues 1054-1066) in Topo IIα
Doubt about NES: Peptide at 1051-1070 failed to bind CRM1 in GST-pulldown assay (Chook lab, unpublished results).
