L5 is part of the 60S ribosomoal subunit. It binds to 5S rRNA and is involved in exporting 5S rRNA from the nucleus to its storage site in the cytoplasm. L5 is predominantly nucleus. The NLSs were mapped to aa 21-35 and 150-297. The proposed NES can direct GST to the cytoplasm.
Doubts about NES: The proposed NES failed to bind CRM1 in GST-pulldown assay (Chook Lab, unpublished results). The export efficiency of the wild type L5 is low. Murdoch et al. showed that although nuclear export of 5S RNP requires CRM1, full length L5 didn't directly bind to CRM1 in vitro. Only the first 25 amino acids. They speculate that maybe complex with 5s rRNA will change conformation of L5.
