X11L2 belongs to a family of highly conserved adaptor proteins which provide molecular scaffolds for membrane proteins and membrane-associated proteins. Some scaffolding proteins have been shown to be involved in the nucleus. Sumioka et al. showed that X11L2 can shuttle and can activate transcription. Efficient shuttling is important for the observed transcriptional activity. Crystal structure of NES peptide bound to CRM1 revealed that X11L2 NES is a class 4 NES which uses a beta turn to accommodate the last leucine into the P4 CRM1 hydrophobic pocket.
