Pericentrin plays a crucial role in organization and function of centrosomes and mitotic spindles, maintenance of cell cycle integrity, and regulation of DNA damage checkpoint. Single-NES mutations do not change pericentrin cytoplasmic localization. Mutation of entire NES, on the other hand, increased accumulation in the nucleus and lead to the proposal that greater number of NES could enhance interactions between CRM1. Associated proteins such as dynein and PKC are capable of retaining pericentrin in the cytoplasm even in the absence of NES.
