Of the four diacylglycerol kinases (DGK) isozymes found in the nucleus, DGKζ is the only one to contain a nuclear localization signal. DGKζ regulates nuclear diacylglycerol levels as well as cell cycle progression from G1 to S phase through decreased phosphorylation of retinoblastoma protein (pRb). Impaired nuclear export of DGKζ increased cell cycle arrent in C2C12 cells. It has also been speculated that the absence of the COOH-terminus of DGKζ enhance its interactions with CRM1.
