Breast cancer susceptibility gene BRCA2 plays a key role in breast and ovarian cancer predisposition. Although it predominantly localizes in the nucleas and plays significant roles in DNA double-strand break repair, it has been known be involved in nucleo-cytoplasmic shuttling. In specific, BRCA2 contains a centrosomal localization signal and is found in the centrosomes during the S and early M phases of the cell cycle. The BRCA2 NES contributes to its centrosomal localization since the protein exists first in the nucleus before translocation. Results suggest that disruption of the NES function by genetic changes results in deregulation of BRCA2 export, which ultimately leads to centrosome disorder.
