Comments
ChREBP controls expression of the carbohydrate response element-containing genes. These genes include liver pyruvate kinase and all liver lipogenic enzymes. It has a NLS, two NESs, a DNA-binding bHLH/Zip domain, and proline-rich domain. The N-terminus of ChREBP interacts with 14-3-3 protein, which can regulate its localization. In the presence of increased circulating glucose, ChREBP is shifted to the nucleus from the cytoplasm, which in turn decreases regulation of LPK expression and lipogenic genes. When exposed to low-glucose conditions, the transcriptional activity of ChREBP is stopped by phosphorylation-mediated protein kinases such as PKA, followed by interaction with 14-3-3. The ChREBP-14-3-3 complex subsequently binds to CRM1 and is localized to the cytosol. Although not proven sufficient, a purported NES is located at amino acids 5-15 as well as 86-95. Both NESs seem to be required for maximum interaction with CRM1.
