ALX plays a role in CD28 mediated up-regulation of the IL-2 promoter, which is a key event in T cell activation in the immune response. ALX is located in both the nucleus and the cytoplasm and nuclear pool of ALX is rapidly depleted in response to TCR and CD28 signaling. Shapiro et al. found a NES located at the C-terminal is responsible for the removal of ALX from the nucleus. The C-terminal also contain sequences for nuclear entry, although no apparent NLS was found.
