Comments
α-actinin is a cross-linking protein that bundles actin filaments. It resembles the spectrin superfamily with its four spectrin repeats. There exist four isoforms of α-actinin. Actinin-2 and actinic-3 are muscle cells specific, while actinin-1 and actinin-4 are ubiquitous. Despite the high sequence similarity, actinin-1 and actinin-4 behave differently. In particular, actinin-4 localizes to the nucleus upon inhibition of phosphoinositide 3-kinase or actin depolymerization in serval cancer cell lines. Kumeta et al. demonstrated that actinic-4 is a nucleocytoplasmic shuttling protein. The export process is CRM1-dependent. Since actinin-4 (152-758) is nuclear and actinin-4 (141-758) is cytoplasmic, the functional NES is deemed to be aa 141-151. Interestingly, the import of actinin-4 is independent of importins. Kumeta et al. proposed that the inner spectrin repeats mediates the import through direct interaction with nuclear pore.
