β-Arrestins are non-visual arrestins involved in the termination of G protein-coupled receptor signaling. β-Arrestins have two isoforms: β-Arrestin-1 and β-Arrestin-2. Despite of the sequence and structural similarity, β-Arrestin-1 and β-Arrestin-2 display distinct subcellular localization. β-Arrestin-1 is found throughout the cell, while β-Arrestin-2 is mainly cytoplasmic. Scott et al. demonstrated that β-Arrestin-2 is a nucleocytoplasmic shuttling protein with one CRM1-dependent NES at the C-terminus. Mutation at either L385 or L397 abolishes export, while mutation of I387 has no effect. Note that the PDBs listed are for β-Arrestin-1, where the single substitution of L395 to Q394 explains the lack of functional NES in β-Arrestin-1. They also showed that the shuttling of β-Arrestin-2 serves to redirect nuclear JNK3 to cytoplasm. Wang et al. reported the NES of β-Arrestin-2 in human (Uniprot: P32121).
