Comments
GEN1 is a structure-selective endonuclease that resolve holiday junctions. Chan and West found GEN1 to be mainly cytoplasmic in interphase cells with or without DNA damage stimuli. This localization is LMB-sensitive only in G1 cells, as well as telophase, when nuclear export is proposed to happen. Truncation analysis revealed fragment 526-683 to have export activity, with putative NES at 660-669, which matches traditional NES consensus. This is in contrast with its yeast ortholog Yen1 which contains an NLS instead, illustrating two different ways of achieving spatiotemporal regulation of their mitotic function.
