RapGEF1 is a Guanine nucleotide-releasing protein for the Ras family GTPases. It is primarily cytoplasmic, but Shakyawar et al. showed that it can be found in the nucleus and identified two NLSs. Deletion analysis mapped an NES to the N-terminal portion of the catalytic domain. The full-length protein is only weakly responsive to LMB, but the moderate change can be abrogated by the dual leucine mutation in the putative NES. The peptide itself can direct GFP's nuclear exit efficiently in a LMB-sensitive manner, however the sequence contains an excess of hydrophobic residues uncharacteristic of functional NESs. Homology model based on RAS guanyl-releasing protein 1 showed the putative NES as a helix in a double helical pair, possibly inaccessible to CRM1.
