Gwl (Mastl in mammals, entry #259) is a Cyclin B/Cdk1 activated kinase, which phosphorylates downstream proteins to regulate mitotic entry and progression. Wang et al. found that Drosophila Gwl is also shuttled to the cytosol in prophase in LMB-sensitive manner. In addition to two putative NLSs, a putative NES can be found in 553-558 (not the same as the ones tested in human Gwl). Kinase dead mutants K87R and S840A have abolished nuclear export activity. NES mutations that affected nuclear export did not affect kinase activity. This study also showed that phosphorylation of 562 and inactivation of the nearby NLS is required for nuclear exclusion.
