USP12 is a member of the human deubiquitinase family. Jahan et al. found USP12 to be exported to the cytosol upon T-cell receptor stimulation. This can be abolished by LMB treatment or mutations to a putative NES in residue 77-99. This region matched non-canonical class 3 NES consensus. García-Santisteban et al. also tested another putative NES in 248-262 in isolation fused to a fluorescence reporter and found no export activity. Olazabal-Herrero et al. showed LMB sensitivity of USP12 localization, but the previously identified NES from Jahan et al. was not active in a nuclear export assay in a Rev(1.4)-GFP reporter. Mutations to the NES region instead disrupted interaction with WDR20 which the authors found to have a functional NES instead (entry #404).
