Comments
TBLR1/TBL1XR1 is a transcription coactivator for nuclear receptors. Daniels et al. found that this protein can be found in mislocalized to the cytoplasm in disease and also found a cytoplasmic isoform that lacks the first 89 amino acids. However, truncation analysis leads to conclusion that region between 80-86 is important for cytoplasmic localization., as 1-86 is cytoplasmic but 1-80 is nuclear. A consensus match for only the LXL in the NES consensus can be found in residue 81-83, with possible extra hydrophobic anchors on both sides of the sequence. Deletion of the region however appears to have no affect in localization and LMB sensitivity was not tested to demonstrate CRM1 dependence. It is unclear this is a true CRM1-dependent NES.
