Borna disease virus X protein is a small non-structural protein with diverse functions in viral replication. It is found in the predominantly in the nucleus and mitochondria. The N-terminal 20 residues have been found to contain a NLS as well as having mitochondrial targeting function. Ferré et al. found that CRM1 can be detected in co-IP with X protein, and a putative NES can be found in the same N-terminal region (class 2 and 1a-R can be found between 5-16). Peptide consisting of 5-16 fused to GFP has LMB-sensitive nuclear export activity, and a milder affect is seen when 1-20 is used. The same peptides also display mitochondrial targeting activity with mutations in residues 2-4 affecting its mitochondrial targeting activity.
