FAM21 is a component of the pentameric WASH complex, one of its function is to anchor the complex to endosomes. Deng et al. found that FAM21 interacts with other proteins such as NFκB, which prompted the authors to investigate the nucleocytoplasmic trafficking of FAM21. They found that FAM21 contains putative NLS and NES. Localization of the protein is moderately LMB sensitive. The putative NES is located in 144-153 and matches {L-x(2,3)-{LIVFM]-x(2,3)-L-x-[LI]} consensus. YFP-FAM21(1-356) is cytosolic, and both NES mutations and LMB caused nuclear accumulation. Deletion analysis also point to 1-219 as the fragment that contains export activity.
