PER proteins (1-3) are circadian regulators and play a role in the regulation of circadian rhythm. In mammals, the nuclear localization of PER proteins is controlled by multiple mechanisms. Vielhaber et al. found that mPER1 the first 205 amino acids are required for nuclear localization of mPER1. On the other hand, the region between amino acid 485-495 is required for its cytoplasmic localization. They further showed that aa 411-498, when fused to YFP-NLS, was sufficient to re-localize YFP-NLS from nucleus to cytoplasm (note: the consensus sequence amoung PER proteins for mPER1 is 486-498). Mutations of I493, L496, and L498 to alanines and lysine prevented the cytoplasmic accumulations of YFP-NLS, and the export was also blocked by LMB treatment.
