Comments
TG2 catalyzes the cross-linking of proteins and regulates cell growth and death. It is found predominantly in the cytoplasm. Putative NES was identified in 657-664 within the C-terminal beta-barrel domain, which matches non-canonical NES consensus (class 3 or possibly reverse classes). Interaction with CRM1 was shown with GST-fusion XPO1, and the interaction can be abolished by LMB, however it is unclear if RanGTP was used. This work took care to not truncate the beta barrel domain into fragments and only did mutations from L to Q. The putative NES is in a folded domain but the hydrophobic residues can be seen pointing out and possibly accessible in certain conformations of the protein.
