PER proteins (1-3) are circadian regulators and play a role in the regulation of circadian rhythm. In mammals, the nuclear localization of PER proteins is controlled by multiple mechanisms. Vielhaber et al. found that aa 411-498 of mPER2, when fused to YFP-NLS, was sufficient to re-localize YFP-NLS from nucleus to cytoplasm (note: the consensus sequence amoung PER proteins for mPER1 is 457-469). Mutations of I464, L467, and M469 to alanines and lysine prevented the cytoplasmic accumulations of YFP-NLS, and the export was also blocked by LMB treatment. They also found the single mutation of M469 to lysine greatly impairs the export ability.
