Myosin VI (unconventional) is the only known myosin that moves towards the minus end of actin filaments, with possible roles in transcription in the nucleus. Majewski et al. found that Myosin VI is translocated in the nucleus upon cell stimulation, and the trafficking is sensitive to pharmacological inhibition of both nuclear import (ivermectin) and export (LMB). Putative NES is found in 1012-1016, with only 3 leucines and no known NES consensus match. Moreover, it is located in a putative coiled-coil domain. No further validation of the putative NES was performed.
