DeSI1 (or POST) is a small protein that interacts with UBQLN4 (or UBIN). Hirayama et al. found that UBIN-POST are exported in a LMB-sensitive manner by the action of an NES within POST. The NES identified is sufficient to direct nuclear exit of Venus fusions and binds CRM1 in vitro in a Ran-dependent manner. Mutations to the NES in the full-length protein caused increase nuclear accumulation. This study showed that CRM1 binds to POST which binds to UBIN, and UBIN in turn recognizes K48-linked polyubiquitinated proteins for nuclear export. The NES is located at the C-terminus of the protein and appears exposed and accessible for CRM1 binding in X-ray structure.
