Comments
RGNEF is a guanine exchange factor and an RNA-binding protein implicated in amyotrophic lateral sclerosis. It is predominantly cytoplasmic with some nuclear localization under normal conditions. Tavolieri et al. identified a bipartite NLS in the PH domain necessary and sufficient for nuclear localization, and an overlapping NES within the linker of the NLS. Export activity was apparent and demonstrated to be LMB-sensitive when two lysine mutations were introduced to the putative NLS. Putative NES 1106-1117 matches traditional NES consensus, but contain an excess of hydrophobic residues, which is usually unfavorable for NES activity. This region has also been shown to be form a strand-turn-strand structure within a beta sheet in the PH domain. The domain will have to be partially unfolded to be accessible for CRM1 binding.
