Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) receptor TRAIL-R2 resides at the plasma membrane and binds TRAIL and induce apoptosis or necroptosis. Most known TRAIL-R2 functions occur at the plasma membrane, but most TRAIL-R2 are found in the cytoplasm and nucleus of tumor cells. Intracellular TRAIL-R2 may allow tumor cells to escape TRAIL-induced apoptosis. Cytoplasmic TRAIL-R2 is involved in UPR and nuclear TRAIl-R2 binds to the microprocessor protein to impair maturation of the let-7 miRNA that leads to increased proliferation. TRAIL-R2 shuttles between the nucleus and the cytoplasm. NetNES 1.1 Predictor found 3 putative NESs in TRAIL-R2. IP TRAIL-R2 and found CRM1 by western blot. LMB increased nuclear TRAIL-R2. Mutation of residues T408, E411 and L413 to alanines also increased nuclear TRAIL-R2. It is not clear how the authors decided to mutate these residues when they found 3 putative NESs. Binding to CRM1 was illustrated by co-IP only.
