NPAS4 is class I bHLH- PAS transcription factor. Previous work has shown that the protein is exclusively nuclear, but Greb-Markiewicz et al. showed that the protein shuttles in COS-7 and Neuro 2a (N2a) cells at 24 hrs before becoming predominantly nuclear after 48 hours. Using truncations of the protein, the authors identified multiple putative NLSs and NESs in the N-terminal bHLH domain, the PAS-2 domain and the C-terminus. Residues 461-580 in the C-terminus also displayed LMB-sensitive export activity that appears to be masked by inclusion of residues 341-460, however no putative NESs was be identified. It should be noted that activity of these mutants in full-length protein was not tested. Regions of NLS and NES detected are highly conserved with human NPAS4.
