EPS8 consists of an N-terminal PTB domain followed by an SH3 domain and a C-terminal effector region. EPS8 regulates Rac-mediated actin remodeling at the cell cortex but also has an NLS and therefore also localizes to the nucleus where it interacts with Forkhead box M1 transcription factor (FOXM1) during G2/M, and is recruited to the CCNB1 and CDC25B promoters. LMB increases nuclear localization of EPS8. The authors used the NES consensus sequence Lx2,3(F/I/L/V/M)x1,2,3Lx(I/V/L and found a partially matching putative NES at amino acids 265RIDRDVQILNHILDDIEFFITKLQ288, which is near a predicted classical-NLS at residues 299 to 309. Both the putative NES and NLS are in a linker that connects the PTB and SH3 domains.
