Comments
PML is a tumor suppressor with seven human isoforms (I-VII), and six of the isoforms (I-VI) contain NLSs. While primarily found in the nucleus, PML-I isoform also contains a putative NES in residues 704-713, which matches traditional NES consensus. The fusion of PML protein with retinoic acid receptor alpha (RARα), which is associated with acute promyelocytic leukemia (APL), results in the disruption in the proper formation of PML nuclear bodies (PML-NBs), which regulate physiological functions in normal cells. Wang et al. showed that PML-I is essential for formation of PML-NBs and LMB treatment or NES deletion can cause nuclear accumulation of PML-I and rescue formation of PML-NBs when co-transfected with PML-RARα. It should be noted that no further testing on the export activity of the putative NES have been performed.
