Comments
Phosphatidylinositol-specific Phospholipase C (PI-PLC) is one of the key enzymes in the intracellular signal transduction pathway. There are several families of PI-PLC. PLC-&delta1; is predominantly located at the plasma memebrance and in the cytosol, while PLC-&delta4 is mostly nuclear. By sequence comparison, Yamaga et al. speculate that aa 164-177 in its EF-hand domain is a typical NES. Mutations of L172A/I174A causes the whole protein to show nuclear presence. They also show the aa 164-177, when conjugated to OV, can cause the fusion protein to be excluded from the nucleus following nuclear injection. The mutant form of NES (L165A/L169A/L172A/I174A/V176A) failed to do so. LMB treatment inhibits the nuclear export of NES-OV. They also showed that LMB induces nuclear accumulation of endogenous PLC-δ1. They also speculated that based on X-ray structure, calcium binding causes conformational change of the EF-hand, exposing the NES sequence for CRM1 to bind.
