Comments
SETDB1 is responsible for the trimethylation of histone H3 to generate the H2K9me3 tag for epigenetic transcriptional repression. Tachibana et al. surprisingly found that over-expressed and endogenous SETDB1 are both predominantly localized to the cytosol, with undetectable levels in the nucleus in various cell lines. LMB caused slight accumulation of the protein in the nucleus but fluorescent signal was very weak. This can be rescued by blockage of proteasome-mediated degradation by MG132, suggesting that SETDB1 which accumulated in the nucleus was being degraded. Putative NES can be found in residues 24-39 and 93-109 using ValidNES and Wregex prediction programs.
