NR4A1 is an orphan nuclear receptor which regulates diverse cellular pathways such as inflammation and differentiation. Katagiri et al. showed that nerve growth factor phosphorylates NR4A1 and induces its nuclear exit which is LMB sensitive. They also identified two putative NLSs in the N-terminal region and three putative NESs in the ligand binding domain (residues 443-452, 502-511 and 555-565). Fragment containing NES2 454-521 showed the strongest export activity amongst different tested. Mutations to the putative NESs separately and together all appear to affect nuclear export of full-length protein. S140 phosphorylation and phosphomimic is sufficient to induce nuclear export. Various papers showed LMB dependency of NR4A1 nuclear export induced by different stimuli, as well as, the need of phosphorylation for NR4A1 nuclear export. Papac-Milicevic et al. showed that NR4A1 can be co-immunoprecipitated with CRM1, partially dependent on presence of ISG12. However, it should be noted that all of the 3 putative NESs are not solvent exposed in the folded structure of ligand binding domain and would require unfolding or dramatic reorganization to be accessible for CRM1.
