Jab1/CSN5 is the fifth component of the COP9 signalosome (CSN) complex. Jab1/CSN5 down-regulate p27Kip1 by directly interact with p27, which was exported to the cytoplasm and induced to be degraded. LMB treatment prevents p27 breakdown, indicating nuclear export plays an important role. Tomoda et al. found that it is Jab1/CSN5 that interacts with CRM1 in vitro binding assays in LMB sensitive manner, and aa 233-242 alone is sufficient for CRM1 binding. Mutations of L237/240/242 to alanines disrupt in vitro binding. However, aa 242-334 can still interact with CRM1. The effect of mutation within NES (233-242) in CRM1 binding is partial. PLEASE NOTE THAT THE FUNCTIONAL NES (233-242) IS ESTABLISHED BY CRM1 BINDING ASSAYS. They also carried out other experiments to show that the N-terminal half of Jab1/CSN5 can form intermediate complex with p27 in proliferating cells.
