Comments
UBR5 is the E3 ubiquitin-protein ligase for the N-end rule pathways. Three putative NESs (classical NES: 2206-2224 and reverse NES: 209-227, 1607-1605) were identified by in silico prediction methods in Sendino et al. using possible XPO1/CRM1-cancer exportome proteins identified previously as putative CRM1-binders by Kirli et al. (not all proteins were validated in full-length). These putative NESs were tested using Rev(1.4)-GFP assay which all showed very weak to no activity. Only 209-227 was tested in SRVB/A reporter with co-expression with CRM1, and showed no change in localization. Therefore, no CRM1-dependency has been demonstrated for any of the NESs.
