Influenza type-D virus (IDV) NS2 is designated as NS2 due to similarity to Influenza A virus (IAV) NEP protein (entry 256) which is implicated in nuclear export of vRNPs. Zhao et al. reported that IDV NS2 accumulate in the cytosol and that it cannot functionally replace IAV virus NS2 in the H1N1 backbone. Through truncation analysis, three peptides were identified as minimal fragment sufficient for directing GFP-fusion nuclear export in an LMB-sensitive manner. Full-length protein also appears to be LMB-sensitive. Mutations that affect export activity was performed in the fragments and not full-length protein. GFP-NES fragments and full-length protein can pull-down CRM1 protein and co-localize in cells. NS2 with alanine mutations to individual NESs and deletion of NES1 and NES2 still retained CRM1 binding activity, and deletion of NES3 is necessary to abolish CRM1 binding.
