MAPK phosphatase (MKP) is a family of dual specificity protein phophsphatases, which dephosphorylate both threonine and tyrosine residues of mitogen-activated protein kinase (MAPK), acting as a form of negative regulation of MAPK activity. Masuda et al. identified a new member of MKP, MKP7, from the gene bank. BAsed on sequence similarity, they speculated that aa 296-313, aa 610-627 are two NLS, while aa 376-385 is a NES. They found that MKP-7 is localized in cytoplasm, but accumulated in the nucleus under LMB treatment, implying that it is a shuttle protein. Deletion of C-terminal (including the proposed NES region) caused the whole protein to be nuclear and insensitive to LMB treatment. They also mentioned that mutation of three Leucines to alanine cause nuclear localization of the protein. But they failed to mention which three (they are four Ls in the proposed NES region).
