Nedd4 is a E3 ubiquitin ligase enzyme, containing a C2/CaLB domain at the N-terimnal, a WW protein-protein interaction domain, followed by a conserved hect domain, which is responsible for catalytic action. Huacani et al. was working to identify a substrate for Nedd4. They found a proline-rich protein, named hPRTB, is a potential candidate. They showed that hPRTB is a substrate of E3 ligases in virto, and Nedd4 regulates the stability of hPRTB in cells. Since hPRTB colocalizes with splicing factors in nuclear speckles and Nedd4 is mainly cytoplasmic, Nedd4 must be able to enter and exit the nucleus. Nedd4 was reported to contain a bipartite NLS between aa 534-550. Huacani et al. used a series of deletion constructs to map the sequence responsible for its cytoplasmic localization. They found that aa 393-409 is required. Mutations L405A/I307A significantly increased hte nuclear presence of Nedd4, so does LMB treatment. Note that the sequence in the SwissProt for human Nedd4 is 100 aa longer in the N-terminus than the numbering in the paper.
