Nerve growth factor (NGF) has effect on the apoptosis of nervous systems. p75 nerotrophin receptor (p75NTR) is a cell surface receptor recognized by NGF during NGF-induced apoptosis. NADE (p75NTR-associated cell death executor) is a p75NTR-associated protien that mediates apoptosis in response to NGF. Through mutational analysis, Mukai et al. identified aa 72-112 are essential for NGF-dependent regulation of apoptosis. Interestingly, they also found aa 41-71 are sufficient to induce apoptosis without p75NTR. Wild-type NADE is localized exclusively in the cytoplasm. Mutations of L94A/L97A/L99A lead to a whole cell distribution. Note that single mutation L99A has no effect on its cellular distribution. In another paper, Mukai et al. 2000, found that L94A/L97A also resulted in nuclear retention. Taken together, I think only L94 and L97 are important. LMB treatment of cells containing wild type NADE also cause diffusely distribution of NADE. In addition, they found that NES motif is also crucial for self-association and NGF dependent apoptosis.
