Comments
STAT3 is one of the seven members of the STATs family. It plays complex roles in proliferation, survival, and development. STAT3 posses important function within the N-terminus and shuttles between the nucleus and the cytoplasm in unstimulated cells in a LMB-sensitive manner. It was proposed that out of the three NESs, NES(404-414) and NES(524-535) regulate the basal export, whereas NES(306-318) plays an important role in postreceptor-simulated export. Stat3 shares >50% sequence identity with Stat1. All three NESs from Stat3 are conserved in Stat1. Note that NES(524-535) in Stat3 has not been verified in STAT1. But it is accessible. In addition, aa 197-205 in Stat1 was proposed to be a NES. The last hydrophobic residues don't align well. Plus, McBride et al. showed that aa 1-369 of Stat1 don't bind CRM1.
