FKHRL1 belongs to the human Forkhead family, which are proapoptotic transcription factors related to embryogenesis, differentiation, and tumorigenesis. In the absence of growth factor, FKHRL1 is present in the nucleus in an unphosphorylated state. Upon growth factor stimulation, FKHRL1 is phosphorylated by protein kinase Akt at T32 and S253 and exported from the nucleus to the cytoplasm, resulting in the inhibition of target gene transcription. Brunet et al. showed that this relocalization was inhibited by LMB treatment. Two kinds of mutations also prevented FKHRL1 export. One is the mutation of critical hydrophobic in the two putative NES sequences (aa 369-378, aa 386-396). This mutant remains localized in the nucleus even in the presence of growth factor, although they still bind to 14-3-3. The other mutant was generated by substituting the key 14-3-3 binding residues T32 and S253 with alanine. Therefore, both NES and 14-3-3 binding are required for export activity of FKHRL1. The nuclear localization of FKHRL1 is due to a putative NLS at aa 248-250 and 269-271. Mutating the arginines to alanine resulted in cytoplasmic localization even in the absence of growth factor. Binding of 14-3-3 protein to FKHRL1 may mask the NLS and prevent re-entry of the phosphorylated FKHRL1 into the nucleus. They further showed that the phophorylation of FKHRL1 occurs in the nucleus and 14-3-3 also plays role in the export in addition to cytoplasmic retention.
