Nrf2 is the most potent member of the three NF-E2-related factors. Nrf proteins heterodimerize with Jun, small Maf, and other proteins and bind to the antioxidant response element in over 100 genes in response to antioxidant stimulants. Nrf2 of mouse shares high sequence similarity with Nrf2 of human. C-terminal identified in human Nrf2 is conserved in mouse Nrf2. Jain et al. also showed that phosphorylation of Tyrosine 568 is required for interaction with CRM1 and export of Nrf2. In addition, Joo et al. showed that AMPK phosphorylates Serine 550 in the NES (conserved in human form as Ser558) which causes nuclear accumulation independent of the action of Leptomycin B.
