Comments
Pap1 is a fission yeast AP-1-like transcription factor, which is involved in oxidative stress response. Its bZip and cysteine-rich domains (CRD) are homologous to Yap1/Yap2. Kudo et al. found that residues 515-533 within CRD are sufficient to export GST-GFP to the cytoplasm. LMB mislocalizes the fusion protein. Single mutations I519A, L522A, L526A, and C532S greatly reduce export activity while F517A has a lesser impact. They also noted that the CRD can export GFP-GST fusion protein in mammmalian cells. In both S. pombe and mammalian cell, DEM ihibits nuclear export of Pap1 in the oxidative stress response possibly due through oxidation C532, and C523.
