NF-ATc1 is one of the four members of the nuclear factor of activated T (NFAT) cells gene family involved in the activation of early response genes. The activity of NFATc is controlled by Ca2+-calcineurin. In resting cells, the N-terminus of NFATc is located in the cytoplasm and phosphrylated at multiple serines. In response to Ca2+ signals, the dephosphorylation by calcineurin unmasked the NLS in NFACT, resulting in nuclear accumulation of NFACT. Upon termination of Ca2+ signaling, NFACT exits the nucleus rapidly, supposedly through CRM1-dependent pathway. Note that overexpression of GSK3, which phosphorylates the serines can enhance NFATc export. On the other hand, lithium, which inhibits GSK3 activity, can reduce export.
