Topoisomerase II relieves DNA torsional stress in replication and transcription. Topoisomerase II-beta is one of two isoforms in humans. The beta isoform is widely expressed and important for differentiation and maturation. Topoisomerase II-beta is a nuclear protein that is excluded from interphase nucleoli. Mirski et al (BBRC, 2003) tested 7 potential NES sequences in Topoisomerase II-beta (segments starting at residues 97, 143, 484, 523, 1034, 1071 and 1215) and found that only the 1034-1044 segment could target GST-GFP out of the nucleus. Mirski et al 2007 showed that Topoisomerase II-beta does not shuttle between the nucleus and the cytoplasm in proliferating cells (heterokaryon shuttling experiments).
